Fire crews were called to tackle a blaze at the derelict Rescobie House Hotel in Leslie.
Scottish Fire and Rescue Service sent three appliances to the scene at around 8pm on Thursday and crews arrived to find the roof of the structure well alight.
The blaze was brought under control just before 9am.
It is not known what caused the fire.
The building was previously gutted by fire in April 2017, amid reports that trespassers had been seen loitering in the vicinity.
In August the same year, a planning application was lodged to have the listed building demolished.
The demolition plans were later withdrawn.
Once a renowned restaurant and hotel, Rescobie sustained significant damage in the 2017 inferno, which at one point required six fire appliances to bring the blaze under control.
