The A92 trunk road near Glenrothes will see £135,000 of improvements to a 500-metre stretch of carriageway between the Cadham and Balfarg junctions.
The works being undertaken by BEAR Scotland on behalf of Transport Scotland will involve the resurfacing of both northbound and southbound lanes.
Works were due to start on Wednesday for seven nights, excluding the weekend.
For safety reasons, the A92 will be closed each night between 7.30pm and 6.30am. Diversion routes will be in place during the road closures.
Andy Thompson, BEAR Scotland’s north east unit representative, said: “These works will repair a number of road surface defects, which will improve the general condition of the road for years to come.
“The works are being carried out overnight to minimise delays. We would, however, encourage motorists to plan their journeys in advance and allow some extra time for travel.”
